As far as I know, all the road going Diablos are 5-speed. As this was one of the latest Diablos released, maybe Lamborghini did not want to release it as a 6-speed cause the Murcielago was already to be released.

why does it matter if the car is a 5 speed manual or a 6. the car is still fast n e way.

this could be do to strength related issues, lightness, or as the other guy said, the 6 speed is on the new Lamborghini
Think of a car with The top speed or a Mclaren, handling of a Lotus, acceleration of a Dauer, and comfort of a Rolls Royce. Ahhh i could only dream
